Basic Principles
Principle of real-time quantitative PCR instrument: Polymerase chain reaction (PCR) is a molecular biology technique used to amplify specific DNA fragments. It can be regarded as a special DNA amplification outside the organism. It consists of several steps including high-temperature denaturation, low-temperature annealing, and moderate-temperature extension, forming a temperature cycle. This allows the target gene to be rapidly amplified. It is characterized by strong specificity, high sensitivity, simple operation, and time-saving, and represents a major innovation in gene amplification technology. PCR technology can specifically amplify extremely small amounts of target DNA by millions of times, greatly enhancing the ability to analyze and detect DNA molecules.
Real-time quantitative PCR technology incorporates corresponding fluorescent dyes or fluorescently labeled probes based on conventional PCR. During the PCR reaction, it monitors the entire PCR process in real-time through changes in fluorescent signals. This method uses fluorescent chemicals to monitor the total amount of products after each polymerase chain reaction (PCR) cycle, and employs internal or external reference methods to quantitatively analyze specific DNA sequences in the test sample.
Basic Structure
The real-time quantitative PCR system is an instrument for real-time detection of reactions, primarily consisting of a gene amplification thermal cycling system, a real-time fluorescence detection system, a microcircuit control system, and a computer and application software. It includes two core functional modules: the thermal cycling system and the real-time fluorescence detection system. The gene amplification thermal cycling system operates on a principle similar to that of traditional gene amplifiers, utilizing semiconductor heating and cooling to complete the thermal cycling process. The fluorescence detection system mainly consists of a fluorescence excitation component, a light signal transmission component, a fluorescence detection component, and a control system.
